Show moreProsocial behavior and empathy are important aspects of developing social relations in childhood. Prior studies showed protracted structural development of social brain regions associated with prosocial behavior. However, it remains unknown how structure of the social brain is influenced by genetic or environmental factors, and whether overlapping heritability factors explain covariance in structure of the social brain and behavior. The current study examined this hypothesis in a twin sample (aged 7-9-year; N = 512). Bilateral measures of surface area and cortical thickness of the medial prefrontal cortex (mPFC), temporo-parietal junction (TPJ), posterior superior temporal sulcus (pSTS), and precuneus were analyzed. Results showed genetic contributions to surface area and cortical thickness for all brain regions. We found additional shared environmental influences for TPJ, suggesting that this region might be relatively more sensitive to social experiences. Genetic factors also influenced parent-reported prosocial behavior (A = 45%) and empathy (A = 59%). We provided initial evidence that the precuneus shares genetically determined variance with empathy, suggesting a possible small genetic overlap (9%) in brain structure and empathy. These findings show that structure of the social brain and empathy are driven by a combination of genetic and environmental factors, with some factors overlapping for brain structure and behavior. In addition, brain research indicated hyper-responsiveness and sustained activation instead of habituation of amygdala activation to emotional faces in both groups. Little is known, however, about whether the same patterns of amygdala habituation are present in these two groups. The current study examined habituation patterns of amygdala activity to emotional faces (fearful, happy and neutral) in adolescents with a DSM-IV depressive and/or anxiety disorder (N = 25), adolescents with CSA-related PTSD (N = 19) and healthy controls (N = 26). Behaviourally, the adolescents from the internalizing and CSA-related PTSD group reported more anxiety to fearful and neutral faces than adolescents from the control group and adolescents from the CSA-related PTSD group reacted slower compared to the internalizing group. At the whole brain level, there was a significant interaction between time and group within the left amygdala. Follow-up ROI analysis showed elevated initial activity in the amygdala and rapid habituation in the CSA-related PTSD group compared to the internalizing group. These findings suggest that habituation patterns of amygdala activation provide additional information on problems with emotional face processing. Furthermore, the results suggest there are differences in the underlying neurobiological mechanisms related to emotional face processing for adolescents with internalizing disorders and adolescents with CSA-related PTSD. Show moreFunctions that rely on dorsolateral prefrontal and parietal cortex, including working memory manipulation, are among the latest functions to mature. Yet, several behavioral studies have shown that children may improve on these functions after extensive practice. In this pilot study, we examined whether children would be able to demonstrate increased frontoparietal brain activation after practice. Twelve-year-old children and young adults practiced for 6 weeks with a working memory manipulation task. Before and after practice, functional magnetic resonance imaging data were acquired. Both children and adults demonstrated better performance, lasting at least up to 6 months after the practice period. Before practice, children showed immature frontoparietal activation for manipulation of information in working memory relative to pure maintenance, specifically during the delay period of the task. After practice, the activation differences between children and adults were considerably reduced, suggesting that children may show increased frontoparietal activation if given extensive practice. These preliminary findings argue against the hypothesis that certain brain structures cannot be engaged because of immaturity. Yet, future studies with larger samples should further examine flexibility in the developing brain, and establish what can and cannot be expected of children across school-aged development. Show less
