Show moreThis thesis describes the synthesis and biological evaluation of TLR2/6, TLR4, TLR7/8 and TLR9 ligands, of which the activity can be conditionally controlled. Trans-cyclooctenes are used to shield the ligand's moiety critical for TLR activation, which can be removed fast and quantitatively by the addition of tetrazine derivatives. Photocages are used in parallel for the same purpose, which can be removed by exposure to low-energetic UV-light. A new photocage has been developed that can be decorated on two positions, i.e. it is bifunctional. This photocage has been used to synthesize a proteasome inhibitor-doxorubicin conjugate, in which the photocage bisects the two groups, which induces apoptosis in targeted multiple myeloma cells with an acquired resistance for proteasome inhibitors. The photocage was also used to synthesize a probe that enables targeting and isolation of CD8+ T cells in viable human melanoma and non-squamous cell lung cancer tissues. Show less

Show moreCancer vaccines are aimed at raising an immune responses aganist defined tumor antigens. The formulation of a vaccine has a direct impact on the potency of the immune response generated. Cancer vaccines require particular effort in their formulation, as the immune system has to overcome immune suppression exerted by the tumor. In this thesis, different formulations to enhance cancer vaccine efficacy were explored. In the first part it is shown how chemical conjugation of antigen and adjuvant in form synthetic Toll-like receptor ligands represent a strategy to enahnce vaccine potency. Next, it is presented how the loading of peptide antigens onto dextran nanogel contributes to increase the breadth of the immune response induced. Lastly, a new DNA vaccine containing multiple tumor neoantigens was explored as a platform for personalized cancer vaccines. Together, these studies display how the formulation of cancer vaccines can be modified and optimized to achieve stronger therapeutic efficacy in the clinic. Show less

Show moreMannose-6-phosphate (M6P) is recognized by the mannose-6-phosphate receptor and plays an important role in the transport of cargo to the endosomes, making it an attractive tool to improve endosomal trafficking of vaccines. We describe herein the assembly of peptide antigen conjugates carrying clusters of mannose-6-C-phosphonates (M6Po). The M6Po's are stable M6P mimics that are resistant to cleavage of the phosphate group by endogenous phosphatases. Two different strategies for the incorporation of the M6Po clusters in the conjugate have been developed: the first relies on a "post-assembly" click approach employing an M6Po bearing an alkyne functionality; the second hinges on an M6PoC-glycoside amino acid building block that can be used in solid-phase peptide synthesis. The generated conjugates were further equipped with a TLR7 ligand to stimulate dendritic cell (DC) maturation. While antigen presentation is hindered by the presence of the M6Po clusters, the incorporation of the M6Po clusters leads to increased activation of DCs, thus demonstrating their potential in improving vaccine adjuvanticity by intraendosomally active TLR ligands.
