Show moreCardiovascular diseases are the leading cause of death worldwide, with atherosclerosis as most common underlying pathology. Atherosclerosis is characterized by arterial narrowing due to cholesterol and lipid accumulation. Despite available effective cholesterol lowering medication, considerable risk for recurrent vascular events remains. This residual risk is at least in part explained by high blood lipid levels. The research described in this thesis revealed novel therapeutic strategies that improve lipid metabolism and reduce atherosclerosis development in mice. Inhibition of the endocannabinoid system was found to be an effective strategy, as well as concomitant activation of two incretin hormone receptors, namely those for GIP and GLP1. For combined GIP/GLP1 receptor agonism we additionally showed strongly attenuated hepatic steatosis. We were also able to identify additional targets to attenuate hyperlipidemia by studying the mechanisms underlying the strong day-night rhythm of brown adipose tissue, which is a lipid combusting tissue. In this thesis, I also stress the importance of the choice in animal model when studying lipid-modifying interventions, and describe the development of the software tool RandoMice which can be used to improve the quality of preclinical studies by creating well-balanced experimental groups. Show moreThe aim of the thesis was to develop metabolic analytical platforms for static and dynamic measurements that could answer biological questions for in vitro and in vivo animal models in the area of lipid research. Gene profiling together with the transcriptome and metabolome data was used in combination with the LC/MS analytical platform. In terms of the analytical platforms developed, the focus was on high resolution LC/MS but not limited, as amalgamation with other platforms such as gradient gel electrophoresis (GGE) and fast protein liquid chromatography (FPLC) were explored in more detail to investigate the lipid composition of lipoprotein particles. These analytical strategies were applied to different lipid modulating biological targets as a mean to obtaining a more detailed and characteristic phenotype description directing decisions in drug search during the drug discovery process on the basis of the analytical results obtained. Show moreIn dit promotieonderzoek is zijn de effecten van vetstapeling en ontstekingsreactie tijdens het proces van atherosclerose. We hebben aangetoond dat het ontstekingsremmende eiwit interleukine-9, een stof die door bepaalde immunologische cellen geproduceerd wordt, een remmende werking heeft op het ontstaan van atherosclerose in het algemeen en van vetstapeling in macrofagen in het bijzonder. Aan de andere kant blijkt uit mijn promotieonderzoek dat vetstapeling van macrofagen de gevoeligheid van deze cellen voor ontstekingen beïnvloedt. LPS is in staat om een zeer sterke ontstekingsreactie te stimuleren en om de expressie van verschillende genen die betrokken zijn bij vetstapeling te beïnvloeden. Door gebruik te maken van muizen die geen scavenger receptor BI (SR-BI) tot expressie brengen, hebben we aangetoond dat SR-BI beschermd tegen de door LPS gestimuleerde ontstekingsreactie. Tevens blijkt dat een dieet met een hoog cholesterol gehalte een grote invloed heeft op parenchymcellen in de lever. Voornamelijk FABP5 en vier nieuwe vetzuurbindende eiwitten lijken een belangrijke rol te spelen in de reactie van deze cellen op het dieet. Ook het ontstekingremmende interleukine 10, waarvan bekend is dat het atherosclerose kan remmen en een verlaging van cholesterol in het bloed kan veroorzaken, beïnvloedt vele genen betrokken bij vethuishouding in parenchymcellen van de lever.
