The current understanding of potential unintended (eco)toxicological impacts of nano-formulated pesticides is scarce, in particular with regard to (non-target) aquatic organisms and ecosystems. The present study reports the results of a long-term freshwater mesocosm experiment which assessed responses of individual zooplankton taxa and communities to a novel TiO2-coated nano-formulation of the fungicidecarbendazim. Population- and community trends were assessed and compared in response to the nano-formulation and its constituents applied individually (i.e. nano-sized TiO2, carbendazim) and in combination (i.e. nano-sized TiO2 & carbendazim). Minimal differences were observed between effects induced by the nano-formulation and its active ingredient (i.e. carbendazim) when applied at equivalent nominal test concentrations (4 μg L−1). Nano-sized TiO2 was found to affect zooplankton community trends when applied separately at environmentally realistic concentrations (20 μg L−1 nominal test concentration). However, when nano-sized TiO2 was applied in combination with carbendazim, nano-sized TiO2 was found not to alter effects on community trends induced by carbendazim. The findings of the current study provide an extensive and timely addition to the current body of work available on non-target impacts of nano-formulated pesticides. Show less

Show moreIn the current industrial scenario, chromium (Cr) as a metal is of great importance but poses a major threat to the ecosystem because of its toxicity, but fewer studies have been conducted on its effects and alleviation strategies by using nanoparticles (NPs) and plant growth promoting rhizobacteria (PGPR). Taking into consideration the positive effects of silver⎯nanoparticles (Ag⎯NPs) and (HAS31) rhizobacteria in reducing Cr toxicity in plants, the present study was conducted. A pot experiment was conducted to determine the effects of single and/or combined application of different levels [0 (no Ag⎯NPS), 15 and 30 mM] of Ag⎯NPs and HAS31 [0 (no HAS31), 50 g and 100 g] on Cr accumulation, morpho-physiological and antioxidative defense attributes of barley (Hordeum vulgare L.) exposed to severe Cr stress [0 (without Cr stress), 50 and 100 μM)]. Results from the present study showed that the increasing levels of Cr in the soil significantly (P < 0.05) decreased plant growth and biomass, photosynthetic pigments, gas exchange attributes, sugars, and nutritional contents from the roots and shoots of the plants. In contrast, increasing levels of Cr in the soil significantly (P < 0.05) increased oxidative stress indicators in term of malondialdehyde, hydrogen peroxide, and electrolyte leakage, and also increased organic acid exudation patter in the roots of H. vulgare. Although, the activities of enzymatic antioxidants and the response of their gene expressions in the roots and shoots of the plants and non-enzymatic such as phenolic, flavonoid, ascorbic acid, and anthocyanin contents were increased by increasing the Cr concentration in the soil. The negative impacts of Cr injury were reduced by the application of PGPR (HAS31) and Ag⎯NPs, which increased plant growth and biomass, improved photosynthetic apparatus, antioxidant enzymes, and mineral uptake, as well as diminished the exudation of organic acids and oxidative stress indicators in roots of H. vulgare by decreasing Cr toxicity. Research findings, therefore, suggest that the application of PGPR (HAS31) and Ag⎯NPs can ameliorate Cr toxicity in H. vulgare, resulting in improved plant growth and composition under metal stress, as depicted by balanced exudation of organic acids. Show moreIncreasing knowledge of the impacts of pesticides on soil ecological communities is fundamental to a comprehensive understanding of the functional changes in the global agroecosystem industry. In this study, we examined microbial community shifts in the gut of the soil-dwelling organism Enchytraeus crypticus and functional shifts in the soil microbiome (bacteria and viruses) after 21 d of exposure to difenoconazole, one of the main fungicides in intensified agriculture. Our results demonstrated reduced body weight and increased oxidative stress levels of E. crypticus under difenoconazole treatment. Meanwhile, difenoconazole not only altered the composition and structure of the gut microbial community, but also interfered with the soil-soil fauna microecology stability by impairing the abundance of beneficial bacteria. Using soil metagenomics, we revealed that bacterial genes encoding detoxification and viruses encoding carbon cycle genes exhibited a dependent enrichment in the toxicity of pesticides via metabolism. Taken together, these findings advance the understanding of the ecotoxicological impact of residual difenoconazole on the soil-soil fauna micro-ecology, and the ecological importance of virus-encoded auxiliary metabolic genes under pesticide stress. Show moreWith the rapid development of nanotechnology in agriculture, there is increasing urgency to assess the impacts of nanoparticles (NPs) on the soil environment. This study merged raw high-throughput sequencing (HTS) data sets generated from 365 soil samples to reveal the potential ecological effects of NPs on soil microbial community by means of metadata analysis and machine learning methods. Metadata analysis showed that treatment with nanoparticles did not have a significant impact on the alpha diversity of the microbial community, but significantly altered the beta diversity. Unfortunately, the abundance of several beneficial bacteria, such as Dyella, Methylophilus, Streptomyces, which promote the growth of plants, and improve pathogenic resistance, was reduced under the addition of synthetic nanoparticles. Furthermore, metadata demonstrated that nanoparticles treatment weakened the biosynthesis ability of cofactors, carriers, and vitamins, and enhanced the degradation ability of aromatic compounds, amino acids, etc. This is unfavorable for the performance of soil functions. Besides the soil heterogeneity, machine learning uncovered that a) the exposure time of nanoparticles was the most important factor to reshape the soil microbial community, and b) long-term exposure decreased the diversity of microbial community and the abundance of beneficial bacteria. This study is the first to use a machine learning model and metadata analysis to investigate the relationship between the properties of nanoparticles and the hazards to the soil microbial community from a macro perspective. This guides the rational use of nanoparticles for which the impacts on soil microbiota are minimized. Show less

Consequently, the future co-release of multiple MNPs will inevitably result in the presence of MNP mixtures in the environment. An important question is if the responses of mixtures of MNPs can be dealt with in a similar way as with the responses of biota to mixtures of metal salts. Moreover, natural organic matter (NOM) is an important parameter affecting the behavior and effect of MNPs. Herein, we determined the joint toxicity and accumulation of copper nanoparticles (CuNPs) and zinc oxide nanoparticles (ZnONPs) in Daphnia magna in the absence and presence of Suwannee River natural organic matter (SR-NOM), compared to the joint toxicity and accumulation of corresponding metal salts. The results of toxicity testing showed that the joint toxicity of CuNPs + ZnONPs was greater than the single toxicity of CuNPs or ZnONPs. The joint toxic action of CuNPs + ZnONPs was additive or more-than-additive for D. magna. A similar pattern was found in the toxicity of the mixtures of Cu- and Zn-salts from the literature data. The presence of SR-NOM had no significant impact on the joint toxicity of CuNPs + ZnONPs. The calculated component-specific contribution to overall toxicity indicated that SR-NOM increased the relative contribution of dissolved ions released from the MNPs to the toxicity of the binary mixtures at high-effect concentrations of individual MNPs. Moreover, dissolved Zn-ions released from the ZnONPs were found to dominate the joint toxicity of CuNPs + ZnONPs in the presence of SR-NOM. Furthermore, the results of the accumulation experiment displayed that the presence of SR-NOM significantly enhanced the accumulation of either CuNPs or ZnONPs in D. magna exposed to the MNP mixtures. Among other groups, pelagic fish are likely to be affected by sound from human activities, but so far have received relatively little attention. Offshore wind farms have become numerous and will become even more abundant in the next decades. Wind farms can be interesting to pelagic fish due to food abundance or fisheries restrictions. At the same time, construction of wind farms involves high levels of anthropogenic noise, likely disturbing and/or deterring pelagic fish. Here, we investigated whether bottom-moored echosounders are a suitable tool for studying the effects of impulsive - intermittent, high-intensity - anthropogenic noise on pelagic fish around wind farms and we explored the possible nature of their responses. Three different wind farms along the Dutch and Belgian coast were examined, one with exposure to the passing by of an experimental seismic survey with a full-scale airgun array, one with pile driving activity in an adjacent wind farm construction site and one control site without exposure. Two bottom-moored echosounders were placed in each wind farm and recorded fish presence and behaviour before, during and after the exposures. The echosounders were successful in detecting variation in the number of fish schools and their behaviour. During the seismic survey exposure there were significantly fewer, but more cohesive, schools than before, whereas during pile driving fish swam shallower with more cohesive schools. However, the types and magnitudes of response patterns were also observed at the control site with no impulsive sound exposure. We therefore stress the need for thorough replication beyond single case studies, before we can conclude that impulsive sounds, from either seismic surveys or pile driving, are a disturbing factor for pelagic fish in otherwise attractive habitat around wind farms. Show moreChronic toxicity of copper (Cu) at sublethal levels is associated with ionoregulatory disturbance and oxidative stress. These factors were considered in a toxicokinetic-toxicodynamic model in the present study. The ionoregulatory disturbance was evaluated by the activity of the Na+/K+-ATPase enzyme (NKA), while oxidative stress was presented by lipid peroxidation (LPO) and glutathione-S-transferase (GST) activity. NKA activity was related to the binding of Cu2+ and Na+ to NKA. LPO and GST activity were linked with the simulated concentration of unbound Cu. The model was calibrated using previously reported data and empirical data generated when zebra mussels were exposed to Cu. The model clearly demonstrated that Cu might inhibit NKA activity by reducing the number of functional pump sites and the limited Cu-bound NKA turnover rate. An ordinary differential equation was used to describe the relationship between the simulated concentration of unbound Cu and LPO/GST activity. Although this method could not explain the fluctuations in these biomarkers during the experiment, the measurements were within the confidence interval of estimations. Model simulation consistently shows nonsignificant differences in LPO and GST activity at two exposure levels, similar to the empirical observation. Show less

Show moreTransition metals are widespread in aquatic environments and can be harmful when concentrations exceed thresholds. Especially for fish, an important component of the human diet, low concentrations of transition metals will directly affect their well-being. Different taxa are protected by unified water quality criteria (WQC) thresholds, which rarely consider the ecological status and economic value of different species. There is therefore an urgent need to study taxon-specific sensitivity. The present study established the species sensitivity distributions of nine typical transition metals (Cr, Mn, Fe, Ni, Cu, Zn, Ag, Cd and Hg) for protecting freshwater and seawater fish based on non-parametric kernel density estimation methods, and then derived their acute and chronic HC5-values. The results showed that Ag and Hg have the highest acute toxic potency to fish in freshwater as well as seawater. Compared with marine fish, freshwater fish were more tolerant to acute exposure to Cr, Fe, Ni and Zn, whilst being more sensitive to Ag and Cd. Moreover, edible fish are more sensitive than other fish to these metals in both freshwater and seawater, encouraging more protection of economically valuable fish that may potentially affect human health. The study provides a strong reference for future research on taxon-specific WQC for transition metals. Show less

Marine mammals are considered indicators for marine ecosystem health and many species are of conservation concern due to a multitude of anthropogenic stressors. Marine mammals may be vulnerable to microplastic exposure from the environment, via direct ingestion from sea water, and indirect uptake from their prey. Here we present the first systematic review of literature on microplastics and marine mammals, composing of 30 studies in total. The majority of studies examined the gastrointestinal tracts of beached, bycaught or hunted cetaceans and pinnipeds, and found that microplastics were present in all but one study, and the abundance varied between 0 and 88 particles per animal. Additionally, microplastics in pinniped scats (faeces) were detected in eight out of ten studies, with incidences ranging from 0% of animals to 100%. Our review highlights considerable methodological and reporting deficiencies and differences among papers, making comparisons and extrapolation across studies difficult. We suggest best practices to avoid these issues in future studies. In addition to empirical studies that quantified microplastics in animals and scat, ten studies out of 30 (all focussing on cetaceans) tried to estimate the risk of exposure using two main approaches; i) overlaying microplastic in the environment (water or prey) with cetacean habitat or ii) proposing biological or chemical biomarkers of exposure. We discuss advice and best practices on research into the exposure and impact of microplastics in marine mammals. Metabolic profiling of zebrafish embryos exposed to selected PFAS, specifically including PFOA and two representative PFECA (i.e., GenX and PFO3TDA), enabled elaboration of an integrated model of the metabolic pathways associated with toxicity of these representative PFAS. Alterations of metabolic profiles suggested targeting of hepatocytes (i.e., hepatotoxicity), as well as apparent modulation of neural metabolites, and moreover, were consistent with a previously proposed role of mitochondrial disruption and peroxisome proliferator-activated receptor (PPAR) activation as reflected by dysfunctions of carbohydrate, lipid and amino acid metabolism, and consistent with a previously proposed contribution of PFAS to metabolic syndrome. Taken together, it was generally concluded that toxicity of PFECA is quantitatively and qualitatively similar to PFOA, and these analogs, likewise, represent potential concerns as environmental toxicants. Show moreWhether toxicity of silver nanoparticles (AgNPs) to organisms originates from the nanoparticles themselves or from the dissolved Ag-ions is still debated, with the majority of studies claiming that extracellular release of Ag-ions is the main cause of toxicity. The objective of this study was to determine the contributions of both particles and dissolved ions to toxic responses, and to better understand the underlying mechanisms of toxicity. In addition, the pathways of AgNPs exposure to plants might play an important role and therefore are explicitly studied as well. We systematically assessed the phytotoxicity, internalization, biodistribution, and antioxidant responses in lettuce (Lactuca sativa) following root or foliar exposure to AgNPs and ionic Ag at various concentrations. For each endpoint the relative contribution of the particle-specific versus the ionic form was quantified. The results reveal particle-specific toxicity and uptake of AgNPs in lettuce as the relative contribution of particulate Ag accounted for more than 65% to the overall toxicity and the Ag accumulation in whole plant tissues. In addition, particle toxicity is shown to originate from the accumulation of Ag in plants by blocking nutrient transport, while ion toxicity is likely due to the induction of excess ROS production. Root exposure induced higher toxicity than foliar exposure at comparable exposure levels. Ag was found to be taken up and subsequently translocated from the exposed parts of plants to other portions regardless of the exposure pathway. These findings suggest particle related toxicity, and demonstrate that the accumulation and translocation of silver nanoparticles need to be considered in assessment of environmental risks and of food safety following consumption of plants exposed to AgNPs by humans. Show moreSuitable methods and fit-for-purpose techniques are required to allow characterization of carbon-based nanomaterials (CB-NMs) in complex matrices. In this study, two methods were developed; a method for extraction and characterization of CB-NMs in biological media and a method for fractionation of natural organic matter (NOM) coated CB-NMs in environmental matrices. The former method was developed by extracting carbon nanotubes (CNTs: sized 0.75 × 3000 nm) and nanoplastics (sized 60, 200 and 600 nm) from eggshells and characterizing the extracted CB-NMs in terms of particle size distribution using asymmetrical flow field-flow fractionation (AF4) coupled with multi-angle light scattering (MALS). The latter method was developed using AF4-MALS to fraction NOM-coated CNT (sized 0.75 × 3000 nm) and nanoplastics (sized 60, 200 and 300 nm) in a simulated natural surface water and provide information about the size distribution of the CB-NM-NOM complexes. The developed AF4-MALS method successfully fractioned the CB-NM-NOM complexes based on hydrodynamic size and provided the size distribution of the complexes. The NOM corona did not shift significantly the median size of the CB-NMs. It influenced however the size distribution of the nanoplastics and CNTs. The sample preparation method failed to extract the CNTs (recovery < 20%) from the matrices of the eggshells while being successful for extracting the nanoplastics (recoveries > 60%). The AF4-MALS fractogram showed that the extraction method did not significantly influence the size distribution of the nanoplastics of 60 and 200 nm size, whereas the peak of 600 nm nanoplastics shifted towards a smaller hydrodynamic size. In conclusion, the developed sample preparation method followed by the developed AF4-MALS method can be applied for extraction, separation and characterization of CB-NMs in biological and environmental matrices. Thus, the methods have a high potential to be methods of choice to investigate CB-NMs in future studies. Show less

Show moreEven though microplastics are intensively studied, the focus of the research is mainly on relatively short term effects at high doses. Therefore there is a need to shift the focus toward more realistic, longer-term endpoints. Studies with a range of chemicals have shown that the response of populations often differs from studies in which a single organism is exposed in an individual container (as often described within standard ecotox screening assays). Here we investigate the impact of primary microplastics (1–5 μm in size) on a population of Daphnia magna. We first allowed a stable population of D. magna to develop over 29 d, after which the populations were exposed to microplastics for three weeks (concentrations ranging from 102 to 105 particles mL−1 and a control). We found a significant impact of microplastics on the total population of D. magna, with a reduction in the amount of adult daphnids. Importantly, when expressed as total biomass, exposure to 105 microplastics mL−1 resulted in a 21% reduction in total biomass compared to control. These results indicate that exposure to microplastics can result in significant adverse effects on the population of D. magna, including a reduction in the number of individuals as well as total biomass. Given the importance of D. magna in freshwater food webs, both as a grazer as well as a food source, this can potentially impact the functioning of the ecosystem. Show less
