Show moreObjectiveTo address the variability in prevalence estimates and inconsistencies in potential risk factors for poststroke cognitive impairment (PSCI) using a standardized approach and individual participant data (IPD) from international cohorts in the Stroke and Cognition Consortium (STROKOG) consortium.MethodsWe harmonized data from 13 studies based in 8 countries. Neuropsychological test scores 2 to 6 months after stroke or TIA and appropriate normative data were used to calculate standardized cognitive domain scores. Domain-specific impairment was based on percentile cutoffs from normative groups, and associations between domain scores and risk factors were examined with 1-stage IPD meta-analysis.ResultsIn a combined sample of 3,146 participants admitted to hospital for stroke (97%) or TIA (3%), 44% were impaired in global cognition and 30% to 35% were impaired in individual domains 2 to 6 months after the index event. Diabetes mellitus and a history of stroke were strongly associated with poorer cognitive function after covariate adjustments; hypertension, smoking, and atrial fibrillation had weaker domain-specific associations. While there were no significant differences in domain impairment among ethnoracial groups, some interethnic differences were found in the effects of risk factors on cognition.ConclusionsThis study confirms the high prevalence of PSCI in diverse populations, highlights common risk factors, in particular diabetes mellitus, and points to ethnoracial differences that warrant attention in the development of prevention strategies. Show moreA critical component of the EuroFlow standardization of leukemia/lymphoma immunophenotyping is instrument setup. Initially, the EuroFlow consortium developed a step-by-step standard operating protocol for instrument setup of >= 8-color flow cytometers that were available in 2006, when the EuroFlow activities started. Currently, there are 14 instruments from 9 manufacturers capable of 3-laser excitation and >= 8 color measurements. The specific adaptations required in the instrument set-up to enable them to acquire the standardized 8-color EuroFlow protocols are described here. Overall, all 14 instruments can be fitted with similar violet, blue and red lasers for simultaneous measurements of >= 8 fluorescent dyes. Since individual instruments differ both on their dynamic range (scale) and emission filters, it is not accurate to simply recalculate the target values to different scale, but adjustment of PMT voltages to a given emission filter and fluorochrome, is essential. For this purpose, EuroFlow has developed an approach using Type IIB (spectrally matching) particles to set-up standardized and fully comparable fluorescence measurements, in instruments from different manufacturers, as demonstrated here for the FACSCanto II, and Navios and MACSQuant flow cytometers. Data acquired after such adjustment on any of the tested cytometry platforms could be fully superimposed and therefore analyzed together. The proposed approach can be used to derive target values for any combination of spectrally distinct fluorochromes and any distinct emission filter of any new flow cytometry platform, which enables the measurement of the 8-color EuroFlow panels in a standardized way, by creating superimposable datafiles. Show moreThe transition from Middle to Upper Paleolithic is a major biological and cultural threshold in the construction of our common humanity. Technological and behavioral changes happened simultaneously to a major climatic cooling, which reached its acme with the Heinrich 4 event, forcing the human populations to develop new strategies for the exploitation of their environment. The recent fieldwork at Les Cottés (France) transitional site offers a good opportunity to document subsistence strategies for this period and to provide for the first time high-resolution insights on its evolution. We present the results of the complete zooarchaeological and taphonomic analysis of the transitional sequence, associated with a large regional synthesis of the subsistence strategy evolution during the Middle to Upper Paleolithic. We conclude that, while there is no major change in the hunting strategies, the butchery activities evolved in strict correlation with the development of range weapons. In addition, the demise of carnivore seems to be a consequence of the human pressure on the environment. Our study demonstrates how the faunal component of the environment became a structuring element of the human social organization, being at the base of future cultural evolutions. Show less

Show moreStudying individuals with extreme phenotypes could facilitate the understanding of disease modification by genetic or environmental factors. Our aim was to identify Huntington's disease (HD) patients with extreme symbol digit modality test (SDMT) scores. We first examined in HD the contribution of cognitive measures of the Unified Huntington's Disease Rating Scale (UHDRS) in predicting clinical endpoints. The language-independent SDMT was used to identify patients performing very well or very poorly relative to their CAG and age cohort. We used data from REGISTRY and COHORT observational study participants (5,603 HD participants with CAG repeats above 39 with 13,868 visits) and of 1,006 healthy volunteers (with 2,241 visits), included to identify natural aging and education effects on cognitive measures. Separate Cox proportional hazards models with CAG, age at study entry, education, sex, UHDRS total motor score and cognitive (SDMT, verbal fluency, Stroop tests) scores as covariates were used to predict clinical endpoints. Quantile regression for longitudinal language-independent SDMT data was used for boundary (2.5% and 97.5% quantiles) estimation and extreme score analyses stratified by age, education, and CAG repeat length. Ten percent of HD participants had an extreme SDMT phenotype for at least one visit. In contrast, only about 3% of participants were consistent SDMT extremes at two or more visits. The thresholds for the one-visit and two-visit extremes can be used to classify existing and new individuals. The identification of these phenotype extremes can be useful in the search for disease modifiers. Show less

Show moreThis study uses data extracted from 3D models to compare blade cores from the Châtelperronian and Protoaurignacian stone tool industries. These technocomplexes are at the center of the debate surrounding the interactions between Neanderthals and anatomically modern humans approximately 45 to 40,000 years ago.We created 3D models of lithic cores from the sites of Roc de Combe and Les Cottés using a standardized photogrammetry protocol. We then used data derived from these 3D models to make quantitative comparisons of artifact attributes that have previously been argued to distinguish the two stone tool industries in question. These attributes include the angle between the platform and flaking surfaces, the shape of core cross sections, and the angle between core axes. The conception of this study was not to privilege the use of new technological and statistical approaches over more traditional or qualitative forms of lithic analysis. Rather, our aim was to experiment with using digital tool to develop nuanced, reproducible ways to describe variability in lithic artifacts.Our results support the hypothesis that there is a difference in the angle between core surfaces between these two industries. Our analysis also indicates a difference in the angle between core axes, although we are more cautious in interpreting these results. An elliptical Fourier analysis of core cross section shape was inconclusive. We discuss what archaeological and methodological factors may have contributed to our results, and the roles of both qualitative and quantitative observations in archaeological research. 3D artifact models generated for this study are included as supplemental data and are available for use by other researchers.
