Show moreMetal uptake and distribution in crops have been demonstrated to be highly variable and depending on the metal of interest and the crop type. However, no consensus is reached regarding the primary factor controlling metal uptake in crops. This study thus comparably investigated Hg, As, Zn, Pb, Cd and Cu uptake and distribution in three crops grown in a watershed near a copper mining field located in Yunnan, Southwestern China. The bioconcentration factor (BCF) and translocation factor (TF) were statistically compared for the same metal across different crops. Leafy crops had a stronger propensity to accumulate Hg, As and Zn than fruit crops. The ability of grain crops to accumulate Cd and Cu was much lower than leafy and fruit crops. The three crops all tended not to accumulate Pb in their edible tissues. The DTPA extracted metal concentrations were not statistically correlated with the metal concentrations in crop edible tissues. It is thus not practical to predict metal uptake of Hg, As, Pb and Zn through their available concentrations in soils. The contents of nitrogen and phosphorus, and competing metal ions present in paddy soil decreased the accumulation of Cu and Cd in rice grains. By means of hierarchical cluster analysis, the high accumulation of Zn in the edible tissues of fruit and grain crops was mainly due to dust inputs via phloem transport from leaves. This is why BCF(Zn) was the highest among the six metals for these two crops. For leafy crops, the accumulation of Hg, Cd and Zn in leaves was mainly through soil inputs by roots. Show moreBackground: Osteofibrous dysplasia-like adamantinoma (OFD-AD) and classic adamantinoma (AD) are rare, neoplastic diseases with only limited data supporting current treatment protocols. We believe that our retrospective multicenter cohort study is the largest analysis of patients with adamantinoma to date. The primary purpose of this study was to describe the disease characteristics and evaluate the oncological outcomes. The secondary purpose was to identify risk factors for local recurrence after surgical treatment and propose treatment guidelines. Methods: Three hundred and eighteen confirmed cases of OFD-AD and AD for which primary treatment was carried out between 1985 and 2015 were submitted by 22 tertiary bone tumor centers. Proposed clinical risk factors for local recurrence such as size, type, and margins were analyzed using univariable and multivariate Cox regression analysis. Results: Of the 318 cases, 128 were OFD-AD and 190 were AD. The mean age at diagnosis was 17 years (median, 14.5 years) for OFD-AD and 32 years (median, 28 years) for AD; 53% of the patients were female. The mean tumor size in the OFD-AD and AD groups combined was 7.8 cm, measured histologically. Sixteen percent of the patients sustained a pathological fracture prior to treatment. Local recurrence was recorded in 22% of the OFD-AD cases and 24% of the AD cases. None of the recurrences in the OFD-AD group progressed to AD. Metastatic disease was found in 18% of the AD cases and fatal disease, in 11% of the AD cases. No metastatic or fatal disease was reported in the OFD-AD group. Multivariate Cox regression analysis demonstrated that uncontaminated resection margins (hazard ratio [HR] = 0.164, 95% confidence interval [CI] = 0.092 to 0.290, p < 0.001), pathological fracture (HR = 1.968, 95% CI = 1.076 to 3.600, p = 0.028), and sex (female versus male: HR = 0.535, 95% CI = 0.300 to 0.952, p = 0.033) impacted the risk of local recurrence. Conclusions: OFD-AD and AD are parts of a disease spectrum but should be regarded as different entities. Our results support reclassification of OFD-AD into the intermediate locally aggressive category, based on the local recurrence rate of 22% and absence of metastases. In our study, metastatic disease was restricted to the AD group (an 18% rate). We advocate wide resection with uncontaminated margins including bone and involved periosteum for both OFD-AD and AD. Show moreDuring Agrobacterium tumefaciens-mediated transformation of plant cells a part of the tumour-inducing plasmid, T-DNA, is integrated into the host genome. In addition, a number of virulence proteins are translocated into the host cell. The virulence protein VirE3 binds to the Arabidopsis thaliana pBrp protein, a plant-specific general transcription factor of the TFIIB family. To study a possible role for VirE3 in transcriptional regulation, we stably expressed virE3 in A. thaliana under control of a tamoxifen-inducible promoter. By RNA sequencing we showed that upon expression of virE3 the RNA levels of 607 genes were increased more than three-fold and those of 132 genes decreased more than three-fold. One of the strongly activated genes was that encoding VBF (At1G56250), an F-box protein that may affect the levels of the VirE2 and VIP1 proteins. Using Arabidopsis cell suspension protoplasts we showed that VirE3 stimulates the VBF promoter, especially when co-expressed with pBrp. Although pBrp is localized at the external surface of plastids, co-expression of VirE3 and pBrp in Arabidopsis cell suspension protoplasts resulted in the accumulation of pBrp in the nucleus. Our results suggest that VirE3 affects the transcriptional machinery of the host cell to favour the transformation process. Show less

Show moreAgrobacterium tumefaciens is a gram-negative soil bacterium that induces plant tumors by transferring a segment of DNA, called T-DNA, into plant cells. Under laboratory conditions, Agrobacterium can also transform many different non-plant organisms such as the yeast Saccharomyces cerevisiae. During this process, a number of virulence proteins, including VirF and VirE3, are translocated into the host cell. VirF contains an F-box domain and, according to current theory, in plants and in yeast may induce degradation of the virulence protein VirE2 and the transcription factor VIP1, required for the integration of the T-DNA into host chromosomal DNA. VirE3 functions as a potential plant transcriptional activator. In our study, we expressed the Agrobacterium virulence proteins VirF and VirE3 in Saccharomyces cerevisiae and Arabidopsis thaliana and studied the effect of virF and virE3 expression on the genome-wide transcription in S. cerevisiae and A.thaliana using DNA microarrays and RNA-sequencing.
