Show moreThe northern white rhinoceros (NWR) is probably the earth's most endangered mammal. To rescue the functionally extinct species, we aim to employ induced pluripotent stem cells (iPSCs) to generate gametes and subsequently embryos in vitro. To elucidate the regulation of pluripotency and differentiation of NWR PSCs, we generated iPSCs from a deceased NWR female using episomal reprogramming, and observed surprising similarities to human PSCs. NWR iPSCs exhibit a broad differentiation potency into the three germ layers and trophoblast, and acquire a naïve-like state of pluripotency, which is pivotal to differentiate PSCs into primordial germ cells (PGCs). Naïve culturing conditions induced a similar expression profile of pluripotency related genes in NWR iPSCs and human ESCs. Furthermore, naïve-like NWR iPSCs displayed increased expression of naïve and PGC marker genes, and a higher integration propensity into developing mouse embryos. As the conversion process was aided by ectopic BCL2 expression, and we observed integration of reprogramming factors, the NWR iPSCs presented here are unsuitable for gamete production. However, the gained insights into the developmental potential of both primed and naïve-like NWR iPSCs are fundamental for in future PGC-specification in order to rescue the species from extinction using cryopreserved somatic cells. Show moreWe investigate the relationship between H{$α$} and [O II]({$λ$}3727) emission in faint star-forming galaxies at z = 1.47 with dust uncorrected star formation rates (SFRs) down to 1.4 M$_{⊙}$ yr$^{-1}$, using data in two narrow bands from wide-field camera/United Kingdom Infrared Telescope and Subaru prime focus camera/Subaru. A stacking analysis allows us to investigate H{$α$} emission flux from bright [O II] emitters as well as faint ones for which H{$α$} is not individually detected, and to compare them with a large sample of local galaxies. We find that there is a clear, positive correlation between the average H{$α$} and [O II] luminosities for [O II] emitters at z = 1.47, with its slope being consistent with the local relation. [O II] emitters at z = 1.47 have lower mean observed ratios of H{$α$}/[O II] suggesting a small but systematic offset (at 2.8{$σ$} significance) towards lower values of dust attenuation, A$_{Hα}$ {\tilde} 0.35, than local galaxies. This confirms that [O II] selection tends to pick up galaxies which are significantly less dusty on average than H{$α$}-selected ones, with the difference being higher at z = 1.47 than at z = 0. The discrepancy of the observed line ratios between [O II] emitters at z = 1.47 and the local galaxies may in part be due to the samples having different metallicities. However, we demonstrate that metallicity is unlikely to be the main cause. Therefore, it is important to take into account that the relations for the dust correction which are derived using H{$α$} emitter samples, and frequently used in many studies of high-z galaxies, may overestimate the intrinsic SFRs of [O II]-selected galaxies, and that surveys of [O II] emission galaxies are likely to miss dusty populations. Show moreWe present the first near-IR scattered light detection of the transitional disk associated with the Herbig Ae star MWC 758 using data obtained as part of the Strategic Exploration of Exoplanets and Disks with Subaru, and 1.1 {$μ$}m Hubble Space Telescope/NICMOS data. While submillimeter studies suggested there is a dust-depleted cavity with r = 0.''35, we find scattered light as close as 0.''1 (20-28 AU) from the star, with no visible cavity at H, K', or K$_s$ . We find two small-scaled spiral structures that asymmetrically shadow the outer disk. We model one of the spirals using spiral density wave theory, and derive a disk aspect ratio of h ~{} 0.18, indicating a dynamically warm disk. If the spiral pattern is excited by a perturber, we estimate its mass to be 5$^{+3}$ $_{- 4}$ M$_J$ , in the range where planet filtration models predict accretion continuing onto the star. Using a combination of non-redundant aperture masking data at L' and angular differential imaging with Locally Optimized Combination of Images at K' and K$_s$ , we exclude stellar or massive brown dwarf companions within 300 mas of the Herbig Ae star, and all but planetary mass companions exterior to 0.''5. We reach 5{$σ$} contrasts limiting companions to planetary masses, 3-4 M$_J$ at 1.''0 and 2 M$_J$ at 1.''55, using the COND models. Collectively, these data strengthen the case for MWC 758 already being a young planetary system. Show less
