Show moreWind and solar photovoltaic (PV) power form vital parts of the energy transition toward renewable energy systems. The rapid development of these two renewables represents an enormous infrastructure construction task including both power generation and its associated electrical grid systems, which will generate demand for metal resources. However, most research on material demands has focused on their power generation systems (wind turbines and PV panels), and few have studied the associated electrical grid systems. Here, we estimate the global metal demands for electrical grid systems associated with wind and utility-scale PV power by 2050, using dynamic material flow analysis based on International Energy Agency's energy scenarios and the typical engineering parameters of transmission grids. Results show that the associated electrical grids require large quantities of metals: 27-81 Mt of copper cumulatively, followed by 20-67 Mt of steel and 11-31 Mt of aluminum. Electrical grids built for solar PV have the largest metal demand, followed by offshore and onshore wind. Power cables are the most metal-consuming electrical components compared to substations and transformers. We also discuss the decommissioning issue of electrical grids and their recovery potential. Show moreAreTomo, an abbreviation for Alignment and Reconstruction for Electron Tomography, is a GPU accelerated software package that fully automates motion-corrected marker-free tomographic alignment and reconstruction in a single package. By correcting in-plane rotation, translation, and importantly, the local motion resulting from beam-induced motion from tilt to tilt, AreTomo can produce tomograms with sufficient accuracy to be directly used for subtomogram averaging. Another major application is the on-the-fly reconstruction of tomograms in parallel with tilt series collection to provide users with real-time feedback of sample quality allowing users to make any necessary adjustments of collection parameters. Here, the multiple alignment algorithms implemented in AreTomo are described and the local motions measured on a typical tilt series are analyzed. The residual local motion after correction for global motion was found in the range of +/- 80 angstrom, indicating that the accurate correction of local motion is critical for high-resolution cryo-electron tomography (cryoET). Show moreThe COVID-19 pandemic has increased negative emotions and decreased positive emotions globally. Left unchecked, these emotional changes might have a wide array of adverse impacts. To reduce negative emotions and increase positive emotions, we tested the effectiveness of reappraisal, an emotion-regulation strategy that modifies how one thinks about a situation. Participants from 87 countries and regions (n = 21,644) were randomly assigned to one of two brief reappraisal interventions (reconstrual or repurposing) or one of two control conditions (active or passive). Results revealed that both reappraisal interventions (vesus both control conditions) consistently reduced negative emotions and increased positive emotions across different measures. Reconstrual and repurposing interventions had similar effects. Importantly, planned exploratory analyses indicated that reappraisal interventions did not reduce intentions to practice preventive health behaviours. The findings demonstrate the viability of creating scalable, low-cost interventions for use around the world. Show less

Show moreAutologous, antigen-specific, tolerogenic dendritic cells (tolDCs) are presently assessed to reverse and possibly cure autoimmune diseases such as type 1 diabetes (T1D). Good Manufacturing Practice production and clinical implementation of such cell therapies critically depend on their stability and reproducible production from healthy donors and, more importantly, patient-derived monocytes. Here the authors demonstrate that tolDCs (modulated using 1,25-dihydroxyvitamin D3 and dexamethasone) displayed similar features, including protein, transcriptome and epigenome profiles, between two international clinical centers and between T1D and healthy donors, validating reproducible production. In addition, neither phenotype nor function of tolDCs was affected by repeated stimulation with inflammatory stimuli, underscoring their stability as semi-mature DCs. Furthermore, tolDCs exhibited differential DNA methylation profiles compared with inflammatory mature DCs (mDCs), and this was already largely established prior to maturation, indicating that tolDCs are locked into an immature state. Finally, approximately 80% of differentially expressed known T1D risk genes displayed a corresponding differential DNA methylome in tolDCs versus mDCs and metabolic and immune pathway genes were also differentially methylated and expressed. In summary, tolDCs are reproducible and stable clinical cell products unaffected by the T1D status of donors. The observed stable, semi-mature phenotype and function of tolDCs are exemplified by epigenetic modifications representative of immature-stage cells. Together, the authors' data provide a strong basis for the production and clinical implementation of tolDCs in the treatment of autoimmune diseases such as T1D. Show moreThis paper is one in a series that explores the importance of radius as a second parameter in galaxy evolution. The topic investigated here is the relationship between star formation rate (SFR) and galaxy radius (R-e) for main-sequence star-forming galaxies. The key observational result is that, over a wide range of stellar mass and redshift in both CANDELS and SDSS, there is little correlation between SFR and R-e at fixed stellar mass. The Kennicutt-Schmidt law, or any similar density-related star formation law, then implies that smaller galaxies must have lower gas fractions than larger galaxies (at fixed M-*), and this is supported by observations of gas in local star-forming galaxies. We investigate the implications by adopting the equilibrium "bathtub" model: the ISM gas mass is assumed to be constant over time, and the net SFR is the difference between the accretion rate of gas onto the galaxy from the halo and the outflow rate due to winds. To match the observed null correlation between SFR and radius, the bathtub model requires that smaller galaxies at fixed mass have weaker galactic winds. Our hypothesis is that galaxies are a two-parameter family whose properties are set mainly by halo mass and concentration. These determine the radius and gas accretion rate, which in turn predict how wind strength needs to vary with R-e to keep the SFR constant. Show less

Show moreIntroduction: In an earlier report of the ASCEND-8 study (open-label, phase I, three-arm study, treatment-naive patients and pre-treated patients with advanced/metastatic NSCLC), it was shown that ceritinib 450 mg with food had comparable exposure and better gastrointestinal tolerability than 750-mg fasted.Methods: Here, we report efficacy and updated safety data from primary efficacy analysis of the ASCEND-8 study. Key secondary endpoints were overall response rate and duration of response, assessed by blinded independent review committee (BIRC) using Response Evaluation Criteria in Solid Tumors 1.1.Results: In total, 306 patients were randomized to ceritinib 450-mg fed (n = 108) or 600-mg fed (n = 87) or 750-mg fasted (n = 111), of which 304 patients were included in safety analysis and 198 treatment-naive patients (ALK receptor tyrosine kinase [ALK]-positive by immunohistochemistry) were included in the efficacy analysis (450-mg fed [n = 73], 600-mg fed [n = 51], and 750-mg fasted [n = 74]). The BIRC-assessed overall response rate was 78.1% (95% confidence interval [CI]: 66.9-86.9), 72.5% (95% CI: 58.3-84.1), and 75.7% (95% CI: 64.3-84.9), respectively; and the median duration of response (months) by BIRC was not estimable (NE) (95% CI: 11.2-NE), 20.7 (95% CI: 15.8-NE), and 15.4 (95% CI: 8.3-NE), respectively. Based on the safety analysis (n = 304), the 450-mg fed arm showed the highest median relative dose intensity (100% versus 78.5% versus 83.7%), lowest proportion of patients with dose reductions (24.1% versus 65.1% versus 60.9%), and lowest proportion of patients with gastrointestinal toxicities (75.9% versus 82.6% versus 91.8%).Conclusion: Ceritinib at a dose of 450 mg with food compared to 750-mg fasted showed consistent efficacy and less gastrointestinal toxicity.
