The issue of ageing, and especially of starvation resistance, has been approached from both the environmental and the genetic sides that are of relevance to the issue. Therewith, this study has a broad orientation, varying from ecophysiology to selection experiments, mutant analysis and sate of the art microarray gene expression analysis. All this work was performed on Drosophila melnogaster, the fruit fly, and was put in an integrative context with human ageing. Show less

Show moreDe Europese aal blijkt bij uitstek een langeafstandsspecialist te zijn. Dit is gebleken uit zwemtesten in 22 zwemtunnels. Het kost schieralen slechts 40% van de vetreserves om de 5500-km naar de paaigronden in de Sargassozee te overbruggen, mogelijk zelfs in slechts drie-en-een-halve maand. Niet alleen dienen vetten als brandstof, ze zijn ook essentieel als energievoorraad voor de toekomstige larven. Het blijkt nu dat zwemmen de afzetting van de vetten in de eitjes op gang brengt. Door te zwemmen beginnen de eicellen van, vooral oudere, alen te ontwikkelen zodat ze kunnen afpaaien zodra ze in de Sargasso zijn aangekomen. De zwemblaasparasiet beschadigt echter de zwemblaas waardoor geïnfecteerde alen minder efficiënt zwemmen en al het vet waarschijnlijk is verbruikt voordat ze de Sargasso bereiken. Een andere verstorende factor zijn de in het vet opgeloste PCB's, die al in zeer lage doses de vroege ontwikkeling van embryo's onmogelijk blijken te maken. Deze twee factoren lijken een belangrijke oorzaak te zijn in de sterke afname van de wilde populatie. Het feit dat we met zwemmen een natuurlijke 'trigger' hebben gevonden van ontwikkeling van de eicellen en dat we in staat zijn geweest schieralen in gevangenschap tot reproductie te brengen biedt hoop voor toekomstige kweek. Show moreThis thesis will first document on variation in progeny sex ratios among individual female plants of Urtica dioica at our field site in Meijendel (Chapter 2). Next, we show that there is also considerable sex ratio variation among male and female flowering shoots in 26 natural populations studied (Chapter 3). Additionally, we studied life history traits of male and female clones to detect sex-specific differences that might have contributed to the sex ratio bias observed in the field. Our results indicate that the sex ratio bias in natural populations may be a consequence of a bias that already originated in the primary sex ratio. Next, we investigated physiological, environmental (Chapter 4) and genetic (Chapter 5) aspects of sex determination. For the latter aspect, a series of crosses including male, female and monoecious plants of U. dioica was performed. These experiments were designed to investigate whether sex ratio variation in the primary sex ratio is due to environmental sex determination (ESD) or genetic sex determination (GSD) solely or due to an interaction of both. Genetic mapping of sex-linked markers was used as a supplementary tool to analyze the genetic mechanism of sex determination (Chapter 6). Finally, to study the inheritance pattern of the sex ratio trait, crosses were performed between individual male and female plants from different sex ratio families (Chapter 7). Show less

Show moreThe biocontrol strain studied in this thesis is Pseudomonas chlororaphis strain PCL1391, which controls tomato foot and root rot caused by Fusarium oxysporum f. sp. radicis-licopersici (Chin-A-Woeng et al., 1998). Production of the anti-fungal metabolite phenazine-1-carboxamide (PCN) and an efficient root colonizing ability (Chin-A-Woeng et al., 2000) are crucial for efficient biocontrol by P. chlororaphis strain PCL1391. This thesis aims at the identification and analysis of environmental conditions affecting the production of PCN and the identification of novel genes involved in the regulation of PCN production. Show moreA general overview of regulation of secondary metabolism in Pseudomonas species is given in Chapter 1. Several approaches were combined to identify novel genes involved in the regulation of PCN synthesis and to study their interactions with other regulators. Site-directed mutagenesis was used to test the hypothesis that rpoS is a regulatory gene of PCN synthesis (Chapter 2). To discover additional genes in the regulatory cascade, which already contains psrA and rpoS, a random DNA-fragment microarray of the PCL1391 genome was constructed and used for transcriptomics of the psrA and rpoS mutants (Chapter 3). A random mutagenesis approach resulted in the identification of pip, a novel gene that stimulates PCN production in PCL1391 (Chapter 4). Analyses on the role of Pip as a switch of PCN production depending on environmental conditions are described in Chapter 5. The results described in this thesis are summarized in Chapter 6, where in addition the regulatory network of PCN synthesis in P. chlororaphis PCL1391 is compared to regulatory networks of secondary metabolism in other Pseudomonas species. Show morePlants defend themselves against stress, including pathogen or herbivore attack, via biosynthesis of defense proteins and of protective compounds called secondary metabolites. Stress induces these responses via a complex signal transduction network with jasmonic acid (JA) and related compounds as major players. In some defense responses JA acts synergistically with the stress hormone ethylene. It is largely unknown how JA induces expression of defense genes. It is also not well understood how the synergistic interaction between JA and ethylene is brought about. Previous research has identified several genes encoding AP2/ERF-domain transcription factors, called ORAs, whose expression is rapidly induced by JA in Arabidopsis. In this thesis, the function of several ORAs is addressed and shows that certain ORAs, by regulating the expression of defense-related genes, play major roles in JA signaling and defense. Show less

Show moreEen nieuwe ontwikkeling in de bierbrouwerij is het gebruik van hogere wortconcentraties in een proces dat 'high-gravity brewing' wordt genoemd. Dit proces heeft als voordeel dat het de produktiviteit van de brouwerij verhoogt. Een nadeel is echter dat door de hogere suikerconcentraties de capaciteit van gist om deze suikers om te zetten in alcohol afneemt. In het bijzonder maltotriose, na maltose de meest voorkomende fermenteerbare suiker in wort, wordt als gevolg van de afgenomen capaciteit vaak niet volledig omgezet. De resultaten van dit onderzoek hebben geleid tot nieuwe informatie om de omzetting van maltotriose te verbeteren. Voor het vergisten van maltotriose zijn twee stappen nodig: eerst aktieve opname en vervolgens afbraak in de cel. Het MTT1-gen dat codeert voor een maltotriose transporter werd ge_soleerd en gekarakteriseerd. De introductie van een veranderde versie van dit gen in industriële giststammen verhoogt de maltotriose opname met een factor 22. Verder is gevonden dat als het enzym __-glucosidase dat maltotriose omzet in de gemakkelijk opneembare suikers glucose en maltose aan de buitenkant van de gistcel wordt gezet, dit kan helpen bij een betere vergisting van maltotriose. Beide manieren geven de brouwers de mogelijkheid om gisstammen te ontwikkelen met een betere fermentatie van maltotriose in 'high-gravity brewing'. Show less

Show moreThe behavior of single G-protein coupled receptor molecules were studied with single-molecule microscopy in the plasmamembrane during Dictyostelium discoideum chemotaxis. The mobility of the receptor was different in the anterior and posterior regions of living cells migrating towards the source of chemoattractant. This difference of mobility can account for an amplification of the extracellular signal, necessary for chemotaxis. It was also found that the receptor molecules are internalized. Show less
