Show moreIn the Arabidopsis genome there are three highly conserved homologues of the mammalian 56-kD selenium-binding protein (SBP). To study the function of SBP in this model plant, we used a transgenic approach by constitutively overexpressing and down-regulating the endogenous Atsbp1 gene. In the latter case, we employed both a conventional antisense method and gene silencing by intron-containing hairpin RNAs. Atsbp1-overexpressing and silenced plants were phenotypically normal, under standard growth conditions, when compared with wild type plants. Transgenic plants exhibited different growth responses to exogenously supplied selenite, which correlated with the expression levels of Atsbp1. Plants with increased Atsbp1 transcript levels showed enhanced tolerance to selenite, while plants with reduced levels were more sensitive. Our results indicate that, although Atsbp1 does not play a detectable role in the regulation of developmental processes under normal growth conditions, it appears to be involved in processes controlling tolerance of Arabidopsis to selenium toxicity. Zo luidt het doel dat het huidige kabinet zichzelf, de gehele overheid en zelfs de gehele samenleving stelt in het regeerakkoord 'Meedoen, meer werk, minder regels'. Een slagvaardige overheid en participerende burgers zijn daarbij vereist. Verantwoordelijkheid nemen en dragen door beide partijen is de sleutel tot succes (Hoofdlijnenakkoord voor het kabinet CDA, VVD en D'66, 2003: 1-2). Een ambitieus streven, aangezien deze poging om legitimiteit te verwerven onder de bevolking een grote kans heeft van falen. Show less

Show moreHet 4e lustrum der B.I.L. begon met een verjaardagsfeest in besloten kring. Op 22 mei 2005 was de oprichting van de B.I.L. precies 20 jaar geleden. Dit vierde het bestuur gezamelijk met chinees en studiosport. Dit kleine feestje zou gevolgd worden door een week vol met activiteiten, feesten en leerzame onderdelen. Deze week werd georganiseerd door de lustrumcommissie "Lust&Co". De week mag met recht een toppunt worden genoemd in de 20-jarige bestaansgeschiedenis van de B.I.L. De verjaardag is dan ook gevierd zoals het hoort, compleet met lustrum-merchandising en natuurlijk een lustrum-dagblad. Show less

Dinsdag 23 Augustus, 4 uur s'ochtends stond achter het FSW de Commissie Buitenland samen met de andere 27 deelnemers en onze gids, Suzanne, klaar om te vertrekken, klaar om te beginnen aan de reis naar Moskou.Na iets langer gewacht te hebben dan was gepland, vertrokken we om een uurtje of 5 met de bus richting het vliegveld in Düsseldorf. Na een reis van ongeveer 3 uur lang lekker dommelen in de bus kwamen we aan bij het vliegveld. Show less

Nog voordat Stef Blok, de voorzitter van de parlementaire onderzoekscommissie Integratiebeleid, het rapport Bruggen Bouwen aan de Tweede Kamer kan aanbieden, rollen de eerste persberichten al uit de fax bij de (landelijke) media. De commentaren liegen er niet om, het rapport wordt afgedaan als 'vrijblijvend, naïef, waardeloos en vaag 1 • De meeste politieke partijen verwijzen het rapport direct en zonder bestudering naar de prullenbak. De boosdoener w as de conclusie "dat de integratie van veel allochtonen geheel of gedeeltelijk geslaagd is en dat dit een prestatie van formaat is, zowel van de betreffende allochtone burgers als van de hen ontvangende samenleving"2 • Show less

Show moreAan mij de eer en genoegen om als aankomend commissaris Ondenvijs het verslag van het Eerstejaarsweekend (kortweg EJW) te schrijven. Het aantal inschrijvingen was iets aan de lage kant, maar dat kon niet voorkomen dat het een weekend een groot succes was. De aanwezige eerstejaars gingen in ieder geval goed geïntegreerd de EL CID-week in. Klokslag half elf stonden alle eerstejaars paraat op Leiden CS. Enige tijd later voegde de ouderejaars zich tot het gezelschap en was de groep klaar om te vertrekken naar het station van Bodegraven. Uiteindelijk was het nog een flinke groep: naast eerstejaars was ook het bestuur, het aankomende bestuur, de introductie-commissie en de mentoren mee. Dolle boel dus. Show less

Na maanden van voorbereidingen, een broodnodige crash course conversational English en de laatste inkopen met mijn moeder op de markt (drop en stroopwafels), vertrok ik dan eindelijk richting Vancouver. Het grootste avontuur uit mijn leven sinds het behalen van mijn B-diploma stond op het punt te beginnen: ik ging de keuzeruimte van mijn studie invullen met een semester aan de University of British Columbia (UBC). Show moreFlavodoxin II from Azotobacter vinelandii is a "long-chain" flavodoxin and has one of the lowest E(1) midpoint potentials found within the flavodoxin family. To better understand the relationship between structural features and redox potentials, the oxidized form of the C69A mutant of this flavodoxin was crystallized and its three-dimensional structure determined to a resolution of 2.25 A by molecular replacement. Its overall fold is similar to that of other flavodoxins, with a central five-stranded parallel P-sheet flanked on either side by alpha-helices. An eight-residue insertion, compared with other long-chain flavodoxins, forms a short 310 helix preceding the start of the alpha(3) helix. The flavin mononucleotide (FMN) cofactor is flanked by a leucine on its re face instead of the more conserved tryptophan, resulting in a more solvent-accessible FMN binding site and stabilization of the hydroquirione (hq) state. In particular the absence of a hydrogen bond to the N5 atom of the oxidized FMN was identified, which destabilizes the ox form, as well as an exceptionally large patch of acidic residues in the vicinity of the FMN N1 atom, which destabilizes the hq form. It is also argued that the presence of a Gly at position 58 in the sequence stabilizes the serniquinone (sq) form, as a result, raising the E(2) value in particular. Show less

Show moreNitrite reductase (NiR) is a multicopper protein, with a trimeric structure containing two types of copper site: type I is present in each subunit whereas type 2 is localized at the subunits interface. The paper reports on the thermal behaviour of wild type NiR from Alcaligenes faecalis S-6. The temperature-induced changes of the copper centres are characterized by optical spectroscopy and electron paramagnetic resonance spectroscopy, and by establishing the thermal stability by differential scanning calorimetry. The calorimetric profile of the enzyme shows a single endothermic peak with maximum heat absorption at T-m approximate to 100 degrees C, revealing an exceptional thermal stability. The thermal transition is irreversible and the scan rate dependence of the calorimetric trace indicates that the denaturation of NiR is kinetically controlled. The divergence of the activation energy values determined by different methods is used as a criterion for the inapplicability of the one-step irreversible model. The best fit of the DSC profiles is obtained when the classical Lumry-Eyring model, N double left right arrow U double right arrow F, is considered. The simulation results indicate that the irreversible step prevails on the reversible one. Moreover, it is found that the conformational changes within the type-1 copper environments precede the denaturation of the whole protein.
