Show moreMHC class II antigen presentation by B cells is important to activate CD4+ T cells that stimulate the B cell to produce antibodies. Besides this, disruption of MHC class II antigen presentation could play a role in immune escape by tumor cells. This thesis describes MHC class II antigen presentation by B cells during bacterial infections and leukemia. We show that, in contrast to the current dogma, human B cells are able to phagocytose whole, living Salmonella bacteria via their B cell receptor (BCR). Salmonella survives intracellular and is transported by the B cell to the spleen where Salmonella can infect other cells. Although Salmonella uses the B cell to disseminate and escape the immune system, uptake of Salmonella by B cells also results in a good immune response consisting of specific antibody production, antigen presentation and activation of both CD4+ T helper cells and cytotoxic CD8+ T cells that are able to eliminate infected cells. Many leukemic cells express MHC class II. For AML (acute myeloid leukemia) we show that expression of the self-peptide CLIP could be a measure for immune escape because a high percentage of HLA-DR+/CLIP- blasts correlates with a longer disease-free survival. In B-CLL (chronic lymphocytic leukemia), the percentage of activated CD4+ and CD8+ T cells is enhanced and inversely correlated to CLIP. In addition, transcription of all MHC class II genes is increased and B-CLL patients with high HLA-DOA mRNA levels have a less favorable prognosis. Show less

Show moreIn this paper, two approaches for estimating the generation in which a multi-objective evolutionary algorithm (MOEA) shows statistically significant signs of convergence are introduced. A set-based perspective is taken where convergence is measured by performance indicators. The proposed techniques fulfill the requirements of proper statistical assessment on the one hand and efficient optimisation for real-world problems on the other hand. The first approach accounts for the stochastic nature of the MOEA by repeating the optimisation runs for increasing generation numbers and analysing the performance indicators using statistical tools. This technique results in a very robust offline procedure. Moreover, an online convergence detection method is introduced as well. This method automatically stops the MOEA when either the variance of the performance indicators falls below a specified threshold or a stagnation of their overall trend is detected. Both methods are analysed and compared for two MOEA and on different classes of benchmark functions. It is shown that the methods successfully operate on all stated problems needing less function evaluations while preserving good approximation duality at the same time. Show less

Show moreThis thesis is about weakly driven granular flows and suspensions. Chapter 1 is an overview of the current knowledge of slow granular flows in so-called split-bottom geometries, which in essence consist of a disk rotating at the bottom of a container. In chapter 2 we study dry granular flows in this split-bottom geometry, both in the frictional, slow, rate-independent regime, and in the liquid-like, rate dependent regime which is reached for faster flows. Chapters 3-5 deal with the flow of suspensions in the same geometry. We improve the so-called index matched scanning technique, that allows 3D imaging of the suspensions. Also for the suspension we study both the slow, rate independent and the faster, rate dependent regime. In all cases we combine 2D and 3D imaging of the flow with rheological measurements. Chapter 6 is devoted to the rheology of dry, weakly vibrated granular media. In chapter 7 we revisit a classic experiment on the compaction of granular media. Show less

Show moreHet onderzoek in het proefschrift heeft betrekking op het opzetten van een theoretisch kader voor het modeleren van ziekteprogressie op een mechanistische grondslag. De nadruk ligt op de uitwerking van dit concept voor een chronisch progressieve ziekte; postmenopausale osteoporose. De onderliggende aanname is dat een betere, samenhangende, beschrijving van dit soort ziektes wordt verkregen door de combinatie van 1) een wiskundige structuur gebaseerd op het onderliggende biologische mechanisme met 2) fysiologische data die de ziekteprogressie en de behandelingseffecten weergeven. Het onderzoek heeft aangetoond dat het modeleren van postmenopausale osteoporose op basis van een ziektesysteem analyse (__disease system analysis__) leidt tot waardevolle inzichten in zowel symptomatische als beschermende (ziekte modificerende) effecten op verschillende biologische markers. Er kan gesteld worden dat deze mechanistische ziektesystemen een __kennisbank__ kunnen en moeten vormen om een beter ge_nformeerd besluitvormingsproces tijdens de ontwikkeling (farmaceutische industrie), de beoordeling (overheden) en uiteindelijk het klinische gebruik van geneesmiddelen. Ziekteprogressie modellen maken het steeds beter mogelijk om informatie vanuit meerdere bronnen samen te voegen op basis van de kennis over het geneesmiddel en de ziekte. Met toenemende kennis over het systeem en de behandelingseffecten van bestaande en nieuwe geneesmiddelen kunnen deze modellen continue worden verbeterd en ingezet in het onderzoek en gebruik van deze middelen. Uiteindelijk leidt dit tot een meer effici_nte en kosteneffectieve benadering van geneesmiddelontwikkeling en het klinisch gebruik van geneesmiddelen. Het is van groot belang dat de farmaceutische industrie, academische instellingen en de overheid verder samenwerken aan het onderzoeken en vergroten van de mogelijkheden van een dergelijke gestructureerde aanpak (www.tipharma.nl). Show moreKoptische muziek, de muziek van de christelijke bevolking van Egypte, is in de Westerse Wereld een onbekend en weinig onderzocht gebied. Alleen van liturgische Koptische muziek zijn ongeveer 300 melodieën uit een eeuwenlange mondelinge traditie bewaard gebleven. De eenstemmige muziek wordt als wisselgezang door zowel solisten als koren voorgedragen. Om meer over de structuur van deze onbekende muziek te weten te komen heeft de auteur deels opnames verzameld, deels rituelen zelf opgenomen in verschillende Koptische kloosters en kerken. Als onderwerp van onderzoek werden de nachtelijke gezangen tussen zonsondergang- en opgang gekozen, Psalmodia genaamd. Met meer dan 100 muzikale transcripties zijn deze melodieën voor volgende generaties vastgelegd. De gezangen van beroemde kerkzangers uit verschillende delen van Egypte werden met elkaar vergeleken. Uit de analyses van deze melodieën bleek dat de improvisatorisch lijkende, lange cadensen een duidelijke basismelodie hebben, die in alle interpretaties terug te vinden is. Daarnaast bestaan melodische formules die de zangers in staat stellen de basismelodieën aan de verschillende feestelijke situaties aan te passen. De keuze van deze formules is gedeeltelijk door de traditie vastgelegd, gedeeltelijk past iedere cantor ze aan zijn eigen zangkunst aan. Show moreAccording to a controversial hypothesis, a characteristic unique to human language is recursion. Contradicting this hypothesis, it has been claimed that the starling, one of the two animal species tested for this ability to date, is able to distinguish acoustic stimuli based on the presence or absence of a center-embedded recursive structure. In our experiment we show that another songbird species, the zebra finch, can also discriminate between artificial song stimuli with these structures. Zebra finches are able to generalize this discrimination to new songs constructed using novel elements belonging to the same categories, similar to starlings. However, to demonstrate that this is based on the ability to detect the putative recursive structure, it is critical to test whether the birds can also distinguish songs with the same structure consisting of elements belonging to unfamiliar categories. We performed this test and show that seven out of eight zebra finches failed it. This suggests that the acquired discrimination was based on phonetic rather than syntactic generalization. The eighth bird, however, must have used more abstract, structural cues. Nevertheless, further probe testing showed that the results of this bird, as well as those of others, could be explained by simpler rules than recursive ones. Although our study casts doubts on whether the rules used by starlings and zebra finches really provide evidence for the ability to detect recursion as present in “context-free” syntax, it also provides evidence for abstract learning of vocal structure in a songbird. Show moreBal tashchit is possibly the most important religious principle directly relating to the relationship between man and his environment. This thesis attempts to answer three basic questions: 1) what exactly is bal tashchit? 2) How might a principle of bal tashchit be applied to current environmental problems? 3) What contributions might the principle of bal tashchit make to the world? This thesis examines the classic texts in the Jewish tradition from which bal tashchit is derived, including the source-text (Deuteronomy 20:19-20), which contains important symbolism concerning the relationship between man and his environment. From these texts is developed a principle of bal tashchit which places a great deal of importance on the intellectual and moral-ethical development of man as key factors in the proper management of the natural environment. Included in this principle is a hierarchy for the allocation of resources in a way that will avoid needless destruction, and maximize human welfare in a sustainable way. As a theoretical approach to the management of current environmental problems, the principle of bal tashchit is applied to the recurring water crisis in the land of Israel, and more peripherally, to the problems of climate change and loss of biodiversity. Deze keer is dat alumnus Marco Stehouwer, hij studeerde vanaf 1991 bestuurskunde. Bovendien volgde hij keuzevakken in Rotterdam en hij heeft een half jaar in de Verenigde Staten gestudeerd.
