High-throughput screening using fluorescent cell models for mechanistic evaluation of adverse drug reactions

This thesis aims to test in vitro pipelines to rapidly and cost-effectively identify molecular mechanisms that drive adverse outcomes in clinical use. First, the research explores this retrospectively by screening known FDA approved drugs for the activation of various cellular stress responses. Next, the same strategy is applied on the same model using a new class of drugs, the kinase inhibitors. This is followed by a study on these kinase inhibitors using a human induced pluripotent stem cell derived cell types, to test whether these models are suitable in high-thoughput screening set-ups. Furthermore, the FAIRification of high-content imaging data is discussed and a metadata template is proposed.
